According to studies, Vancomycin minimum inhibitory concentration (MIC) by E-test was 1-2-fold dilutions higher than the MIC by agar dilution and broth dilution. This study was undertaken to find out the susceptibility pattern of vancomycin and to compare agar dilution and E test.Aim: The aim was to compare the susceptibility testing methods in determining vancomycin MIC in MRSA isolates.Methodology: This prospective study was conducted between November 2011 to May 2013 in the Department of Microbiology Vinayaka Missions Kirupananda Variyar Medical College,Salem.A total of 218 MRSA isolated from heterogenous clinical samples received in Microbiology laboratory were stored in nutrient agar vials at 4-10 oC till further processing. Vancomycin MIC was determined by agar dilution, and E -test method, and MIC creep (2µg/ml) was determined. Results: In the present study, of the 218 isolates, 185 were VSSA (MIC 0.5 - 2 µg/ml), 33 isolates were VISA (MIC - 4-8 µg/ml) and no vancomycin resistant S.aureus isolated (MIC >16 µg/ml). The MIC creep (2µg/ml) was 11 %. In the present study, the E test MIC was higher than the agar dilution method.
